You are choosing a port that is not fully configured. You are likely choosing the wrong port and your programming cable is available on a different port. You should use the Windows Device Manager application to determine which COM port you should be choosing in CHIRP.

For example, in my computer the Windows Device Manager application shows a COM1 and COM3 available under the Ports (COM & LPT) branch (see 9-Device Manager.png). Both will show up in CHIRP, however COM1 is assigned to a serial port that is built into my motherboard. Is it internal to the case so nothing is attached to it. Choosing COM1 will result in the same error that you are experiencing (see COM Port Selected Doesn't Exist.png).

The COM port currently assigned to my programming cable is COM3. It is the COM port that I currently have to select in CHIRP to program any radios using this particular programming cable.
